A lonely little dungeon located way out to sea, The Lost Isle is only accessible once your team has a boat. In order to acquire daid boat you'll need to complete one of Partitio's Scent of Commerce quests, specifically the one based in Tropu'hopu. This will allow you to purchase a seafaring vessel of your own for 100,000 leaves.
Once you have your ship, the only way to get to The Lost Isle is via On the Water, the ship exploration map that allows you to travel from port to port. Head to the southeast of the Sundering Sea and you'll find a whirlpool between two landmasses. Approach it and you'll have to fight the Scourge of the Sea, a powerful boss. Defeat it and you can pass through to the waters beyond. The Lost Isle is to the east.

The Lost Isle is a giant maze. Climb the ladder to the top of the maze and you'll see a sign post to the north of you. All it says is 'Discover the true path'. Nice and cryptic.
The majority of what you'll find on The Lost Isle, aside from a plethora of enemies, are treasure chests. They're sitting out in the open, and the paths to them are, for the most part, plainly visible. We'll go through them before getting to The Lost Isle's true treasure. Note that as you explore you'll run into Blood Revenants, and they will occasionally use an attack called Supernova that can, if there are two of them, instantly wipe your whole party. Be prepared for this to happen if you are very unlucky.
(Yes, it happened to me. Twice. Ugh.)
Take the eastern path from the entrance and it will lead you to a chest containing an Ancient Cursed Talisman. Continue to follow this path southeast and it will bring you to a second chest, this containing another Ancient Cursed Talisman. Get on the northern path and make your way west.
Head south once you pass the rear of the sign. On the left side of the entrance you'll come to a chest containing a Blessing in Disguise. (Combine this with a Rosary of Redemption for a solid accessory.) Head back north, then wander to the west and northwest to find a chest containing... another Ancient Cursed Talisman. Starting to see a pattern?
Northeast of here is another chest with an Ancient Cursed Talisman inside. East of this chest the path will split. If you head west from the split you'll eventually find your way to a chest containing a Great Sage's Staff, which makes up for all the talismans. If you go east from the split you'll find your way to a chest containing a Lost Tribe's Dagger. The eastbound path will also curve you back west, if you follow it to the northeast, and eventually bring you to the end of the maze.
... where you'll find another sign. 'Free from wickedness and distraction, Thy mind's eye shall be thy guide.' Say wha? Yep, this place has a secret. Either walk back through the maze or warp out and return to The Lost Isle, then walk up to the first sign, at the south end of the maze. If you look at the floor tiles directly to the left of the sign at night you'll see a faint, blue, westbound path that is invisible during the day. Walk along this...
... and you'll find an Arcanist Descendent at the other end, standing by themselves on a lonely tower. They will give you a Proof of the Arcanist, which unlocks the Arcanist Job. Arcanists are powerful magic users who deal in Light- and Dark-based spells, and can affect combat in a variety of ways. It's a great, advanced Job for spellcasting characters who have blown through the normal assortment of Jobs.
